Eisai Japan Juvela N Tocopherol Nicotinate Soft Gel Capsules (Tocopherol nicotinate) 200 mg: 100 capsules Varicose Veins, Arteriosclerosis

Each capsule contains 100 mg or 200 mg of tocopherol nicotinic acid ester. Contains Yellow No. 5; soft capsules also contain ethyl p-hydroxybenzoate and propyl p-hydroxybenzoate.

C₃₅H₅₃NO₃　Molecular weight 535.80; yellow to orange-yellow liquid or solid; practically insoluble in water; decomposes upon exposure to light.

It has antioxidant, lipid metabolism-improving, microcirculation-stimulating, vasoprotective, and antiplatelet effects; however, the mechanisms of action are not fully understood. It can lower total cholesterol and lipid peroxides while raising HDL levels; it acts directly on vascular smooth muscle to increase blood flow to the auricle; it improves microvascular permeability; it inhibits platelet aggregation induced by epinephrine, arachidonic acid, collagen, and ADP; and it can increase the partial pressure of oxygen in the blood.

Indications:Symptoms associated with hypertension; hyperlipidemia; peripheral circulatory disorders such as atherosclerotic occlusive disease.

The usual daily dose for adults is 300–600 mg of tocopherol nicotinic acid ester, taken orally in three divided doses; the dose may be adjusted based on age and symptoms.
100 mg hard capsules: 3 to 6 capsules per day | 200 mg soft capsules: usually 3 capsules per day.

Single 600 mg dose after a meal: The unchanged drug reached peak concentration in approximately 6 hours (Cmax 0.615 μg/mL, t½ 4.3 h); α-tocopherol reaches peak concentration in approximately 10 hours (Cmax 1.62 μg/mL, t½ 38.5 h). Postprandial Cmax and AUC are approximately 32-fold and 29-fold higher than those observed in the fasted state, respectively; absorption is strongly affected by food intake.

Symptoms associated with hypertension—such as numbness in the hands and feet, dizziness, stiffness in the shoulders and neck, headaches, insomnia, tinnitus, and cold extremities—may improve; thermal imaging of cold extremities in the elderly showed moderate to significant improvement with 60%. After 2 months of treatment for hyperlipidemia, total cholesterol levels decreased, HDL levels increased, and lipid peroxides decreased. It has also demonstrated efficacy in treating peripheral circulatory disorders such as obstructive arteriosclerosis.
